The primary structure of a protein is basically its amino acid sequence. Which amino acid is attached to which amino acid - this is the primary sequence. Methionine-Arginine-Aspartic Acid, etc. This sequence order is determined by the genomic sequence. Whatever the codon on the mRNA is, this will be translated into an amino acid. The next codon will be translated and the amino acid that corresponds to this will be added.
The result is the primary protein sequence is the actual connections of the amino acids, and this order is determined by the DNA sequence (in the exons).
